Well, the second part of the second stanza simply seems to state what the third stanza is for - a clue, or possibly just a red herring, though it has been confirmed as a clue.
The first part of the second stanza is interesting because I think they might go together, "If you like the first or second, you may not like the third,/An ironic definition for the letters that occur." In this case the third would be the definition, and for some reason it would be something you "may" not like if you like either the first or second.
But because of the first stanza, it seems it is necessary to find the first regardless of whether or not you like it. Although it could very well just be referring to the third stanza, which makes sense with the line "So the riddle is reversed" because it was necessary to solve the third stanza first, which would imply the second stanza should be the next solved.
The most interesting part I think is the simple "It is" at the top. Is this in some way a confirmation of the final answer?

The first two words are "it is" and then the it says the riddle is reversed. So reverse "it is" to "is it."
If there is anything certain about this extensive riddle which points to the ironic and ridiculously long word hippopotomonstrosesquipedaliophobia is that it is not short. So if the riddle is "Short Riddle: Is it?" The answer is obviously "no" which is a pretty short word.
